Methods > Cryo-ultramicrotomy

Thin films of biological and material samples may be flash-frozen and examined, or sections of frozen material may be obtained using the Leica EM UC6 Ultramicrotome equipped with an FC6 cryochamber. The cryo-ultramicrotome is available for semi-thin or thin sectioning of frozen specimens. Sections generated in this way can be used in post-embedding staining for the localization of antigens. Antibody penetration into the section is facilitated, producing excellent localization at the light level. At the electron microscopy level, multiple antigens can be detected on a single thin section. This ultramicrotome is also well suited to the sectioning of a variety of material (non-biological) specimens such as polymers.
